Indications

Gestrenol Tablet is used for: Threatened miscarriage, Recurrent miscarriage, Premature labor

Adult Dose

Oral Threatened miscarriage Adult: PO 5 mg 3 times/day for 5-7 days. Recurrent miscarriage Adult: PO 5-10 mg 3 times/day as soon as pregnancy confirmed. Continue treatment at least one month after the critical period. Threatened Premature Labour 40 mg daily until symptoms improve.

Administration May be taken with or without food. May be taken w/ meals if GI upset occurs.

Contra Indications

Hypersensitivity; thrombophleobitis; undiagnosed vag bleeding, incomplete abortion, hormone-dependent carcinoma, cerebral apoplexy, as a diagnostic test for pregnancy; severe hepatic impairment.

Precautions

Discontinue medications if there is sudden partial or complete loss of vision, proptosis or diplopia; migraine and embolic disorders; epilepsy, migraine, asthma, cardiac or renal dysfunction. History of depression, glucose tolerance and diabetic patients. My impair ability to drive operate machinery. Sudden withdrawal of allyloestrenol should be avoided as it leads to intermittent breakthrough bleeding; lactation.

Pregnancy-Lactation

Pregnancy category: Not classified but clinically it is proven that Allylestrenol is safe during pregnancy & has no teratogenic effect on fetus in any trimester.

Interactions

Enhanced clearance with enzyme-inducing drugs eg, carbamazepine, griseofulvin, phenobarbital, phenytoin and rifampicin. Ketoconazole may increase bioavailability of progesterone. Potentially Fatal: Increased ciclosporin concentration.

Adverse Effects

Side effects of Allylestrenol : GI disturbances, appetite/weight change; fluid retention, oedema; acnes, skin rash, urticaria; depression, headache, fever, fatigue; breast changes, hirsutism, changes in libido, altered menstrual cycles or irregular menstrual bleeding (rare).

Mechanism of Action

Allylestrenol is similar to that of natural progesterone. It prepares the uterus for receiving the fertilised ovum and suppresses uterine motility.
